It is understood the 25-year-old will undergo his testing this weekend after City activated the release clause in his contract - a five-year deal signed only last January.
City are believed to have already agreed personal terms with Delph, who shone in his 32 appearances for the struggling Villans last season.
Delph signed for Villa from Leeds in 2009 after making 52 appearances for the Yorkshire club, and returned to Elland Road for a loan spell in 2012 after struggling to break into Villa's first-team.
But he has since gone from strength to strength despite Villa's troubles, scoring eight goals in over 100 appearances for the West Midlands club.
City are keen to add more young English talent, not only to keep in line with the home-grown quota, but also to establish more to their first-team.
They have again made contact with Liverpool for Raheem Sterling, having had two bids of up to £40m rejected.
